Does Biotin Actually Thicken Hair and Nails?
The evidence for biotin supplements improving hair and nail growth in healthy adults is limited, but the safety concerns around lab test interference are very real.

Short answer: Biotin supplements can resolve hair loss and brittle nails caused by a genuine biotin deficiency, and limited evidence suggests a possible benefit for brittle nails even without deficiency. For people who are not deficient, however, the evidence that high-dose biotin thickens hair or accelerates nail growth is not strong enough to draw firm conclusions.
What Is Biotin?
Biotin, also called vitamin B7, is a water-soluble B vitamin. According to the NIH Office of Dietary Supplements (ODS), biotin serves as a cofactor for five carboxylase enzymes: propionyl-CoA carboxylase, pyruvate carboxylase, methylcrotonyl-CoA carboxylase, acetyl-CoA carboxylase 1, and acetyl-CoA carboxylase 2. These enzymes catalyze critical steps in the metabolism of fatty acids, glucose, and amino acids. Biotin also plays a role in histone modifications, which influence how genes are expressed.
Because keratin, the structural protein that makes up hair and nails, depends on amino acid metabolism, it is biologically plausible that biotin status could affect hair and nail quality. Plausibility, however, is not the same as proof of benefit from supplementation in well-nourished adults.
How Much Biotin Do Adults Need?
The NIH ODS reports an Adequate Intake (AI) of 30 micrograms (mcg) per day for adults. No Recommended Dietary Allowance (RDA) has been established because there is not enough data to set one. Most people in the United States meet the AI through food alone. Rich dietary sources include beef liver, eggs, salmon, pork, and sunflower seeds. Gut bacteria also synthesize some biotin, though the contribution to overall status is not precisely quantified.
What Does the Research Actually Show?
Deficiency and Hair Loss: Strong Evidence
The NIH ODS rates as strong the evidence that correcting a biotin deficiency resolves its classic signs, which include a scaly skin rash, hair loss, and neurological symptoms. True biotin deficiency is uncommon in healthy adults eating a varied diet. It is more likely in people with biotinidase deficiency (a rare inherited disorder), those who consume large amounts of raw egg whites over a long period, people receiving long-term parenteral nutrition without biotin, or individuals taking certain anticonvulsant medications (discussed further below).
Supplements in People Without Deficiency: Limited Evidence
The NIH ODS rates as limited the evidence that biotin supplements improve hair, skin, or nail health in people who are not deficient. The studies that exist are generally small, lack control groups, or focus on populations with underlying conditions rather than healthy adults. The ODS similarly rates as limited the evidence that biotin supplementation improves brittle nails specifically. Some small trials have reported improvements in nail thickness and firmness, but the overall body of evidence is not large enough or consistent enough to support a strong conclusion.
In practical terms, this means a supplement may or may not produce noticeable changes in your hair or nails if your biotin status is already adequate. It does not mean the supplement is harmful, only that the benefit has not been demonstrated convincingly in well-designed research.
The Safety Issue You Should Not Ignore
The NIH ODS notes that no Tolerable Upper Intake Level has been established for biotin because it has not been shown to be toxic in humans. High doses ranging from 10 to 50 milligrams (mg) per day, and up to 200 mg per day in biotinidase deficiency, have not produced toxicity in reported cases. Many over-the-counter hair and nail supplements contain doses far above the 30 mcg AI, sometimes reaching 5,000 to 10,000 mcg (5 to 10 mg) or higher.
The most important practical safety concern is not toxicity but laboratory test interference. The NIH ODS rates as strong the evidence that high-dose supplemental biotin can cause clinically significant interference with certain blood tests. Many modern immunoassays use biotin-streptavidin technology, and excess biotin in the bloodstream can disrupt these tests, producing falsely high or falsely low results.
Specific Tests Affected
- Thyroid function tests: Even a single 10 mg dose has been shown to interfere with thyroid function tests taken within 24 hours, producing a falsely decreased thyroid-stimulating hormone result that can mimic thyrotoxicosis. Case reports describe results falsely indicating Graves' disease and severe hyperthyroidism in patients taking 10 to 300 mg per day.
- Troponin and NT-proBNP: Biotin can falsely lower troponin, the blood marker used to diagnose a heart attack, and NT-proBNP, used to assess heart failure. The FDA has reported a patient death linked to a falsely low troponin result attributed to biotin interference.
- Other analytes: Results for hormones and markers such as 25-hydroxyvitamin D can also be affected.
If you take a biotin supplement and need blood work, tell your clinician and the laboratory. Many labs recommend stopping biotin supplementation for at least 24 to 72 hours before certain tests, though the exact washout period depends on the dose and the specific assay.
Drug Interactions
According to the NIH ODS, long-term treatment with certain anticonvulsants, including carbamazepine, primidone, phenytoin, and phenobarbital, is associated with significantly lower serum biotin levels. This appears to occur because these medications increase biotin catabolism and may inhibit intestinal absorption. People taking these medications should talk to their doctor before starting a biotin supplement, as personal dosing decisions depend on individual health history and current medications. Always consult a clinician or pharmacist before combining any supplement with prescription medication.
Who Might Genuinely Benefit?
Based on current evidence, the people most likely to see a meaningful benefit from biotin supplementation are those with a confirmed or suspected deficiency. If you have unexplained hair loss, a skin rash, or neurological symptoms, a clinician can assess your biotin status and other possible causes before recommending supplementation.
Bottom Line
Biotin plays well-established roles in fatty acid, glucose, and amino acid metabolism, and correcting a deficiency reliably resolves deficiency-related hair and nail changes. The evidence that high-dose biotin supplements thicken hair or strengthen nails in people who are already replete is limited, not strong. The lab interference risk, on the other hand, is supported by strong evidence and has real clinical consequences.
