What Are Glucosamine and Chondroitin?

Your body produces both compounds naturally. The largest clinical trial to date was the Glucosamine and Chondroitin Arthritis Intervention Trial, known as GAIT, funded by the National Institutes of Health and published in the New England Journal of Medicine. The trial enrolled over 1,500 adults with knee osteoarthritis and tested glucosamine alone, chondroitin alone, the two combined, celecoxib, and placebo over 24 weeks.

The NIH grades the evidence that glucosamine or chondroitin relieves pain or improves function in knee osteoarthritis as insufficient. That does not mean the supplements definitively do not work for any individual, but it means the overall body of research has not produced consistent, reliable evidence of benefit across study populations.

Slowing Joint Space Narrowing

Some researchers have looked at whether these supplements might slow the structural progression of osteoarthritis, measured by the narrowing of joint space visible on X-rays. The NIH also grades this evidence as insufficient. At this time, there is not enough consistent data to conclude that glucosamine or chondroitin slows structural joint changes in the knee.

Other Joints

The picture is slightly different for other areas. The NIH grades the evidence that chondroitin helps hand osteoarthritis as limited, meaning there is some signal but not enough to draw firm conclusions. Similarly, the evidence that glucosamine helps hip osteoarthritis is graded limited. These findings are worth noting, but limited evidence still calls for caution before drawing conclusions.

Safety Profile

One reassuring finding from GAIT and other large studies is that no major safety problems have been identified with glucosamine and chondroitin. Adverse events in those trials were mild, infrequent, and distributed similarly between the supplement and placebo groups, according to the NIH Office of Dietary Supplements.

That said, two safety considerations deserve attention for specific groups of people.

Blood Glucose
