High-Dose Zinc and the Risk of Copper Depletion
Taking zinc well above the recommended daily amount can quietly deplete copper, an essential mineral your body needs for energy production and iron metabolism.

Zinc supplements are widely used for immune support and cold relief, but taking doses well above the recommended amount for weeks at a time can reduce your body's copper status. Understanding where the risk begins, and how the two minerals interact, helps you make more informed choices alongside your healthcare provider.
What Zinc Does in the Body
According to the NIH Office of Dietary Supplements (ODS), zinc is required for the catalytic activity of hundreds of enzymes and plays essential roles in immune function, protein and DNA synthesis, wound healing, and cell division. The evidence for these physiological roles is graded Strong by the ODS.
The adult Recommended Dietary Allowance for zinc is 11 mg per day for men and 8 mg per day for women, based on Dietary Reference Intake values from the National Academies. Most people in the United States consume adequate amounts of zinc through food alone.
The Tolerable Upper Intake Level for Zinc
The ODS sets the Tolerable Upper Intake Level (UL) for zinc at 40 mg per day for adults, combining both food and supplement sources. Importantly, this limit was established specifically because higher intakes adversely affect copper status, not because of direct zinc toxicity at that level. The UL does not apply to individuals receiving zinc for medical treatment under the direct care of a physician.
Doses of 50 mg or more per day over several weeks can reduce copper status and may impair immune function, according to ODS safety data. This is a meaningful threshold to keep in mind when evaluating high-potency zinc products on the market.
What Copper Does in the Body
Copper is an essential mineral that serves as a cofactor for cuproenzymes involved in energy (ATP) production, supports iron metabolism largely through the copper-containing enzyme ceruloplasmin, and contributes to the formation of connective tissue. Copper also contributes to antioxidant defense via copper-containing superoxide dismutases. The ODS grades both of these physiological roles as Strong evidence.
The adult Recommended Dietary Allowance for copper is 900 mcg per day, with a Tolerable Upper Intake Level of 10,000 mcg (10 mg) per day. Excessive copper intake can cause gastrointestinal symptoms and, over time, liver damage, so staying within the UL matters in both directions.
How High-Dose Zinc Depletes Copper
The interaction between zinc and copper is well established. High dietary or supplemental zinc interferes with copper absorption in the gastrointestinal tract. According to ODS copper data, moderately high zinc intakes of about 60 mg per day for up to 10 weeks reduced erythrocyte copper-zinc superoxide dismutase, which is a recognized marker of copper status. Excessive zinc use can ultimately cause copper deficiency. The ODS grades the evidence that high-dose zinc supplementation can induce copper deficiency as Moderate.
One practical complication is that copper status is not routinely assessed in clinical settings, and no single biomarker reliably measures it. This means depletion can develop gradually without obvious symptoms until it becomes clinically significant.
Evidence for Common Zinc Uses
It is worth placing the risk in context by reviewing what the research actually supports for zinc supplementation:
- Common cold duration: Supplemental zinc in lozenge or syrup form taken shortly after cold onset may reduce the duration, but not the severity, of common cold symptoms. Evidence is graded Limited.
- Age-related macular degeneration (AMD): AREDS-type supplements containing zinc plus copper and antioxidants slow progression of AMD in people at high risk of the advanced form. Evidence is graded Moderate. Note that these formulations intentionally include copper to offset the zinc-copper interaction.
- Diarrhea in children: Zinc supplementation reduces the duration of acute diarrhea in children in low-income countries. Evidence is graded Moderate.
- Pneumonia in children: Zinc supplementation reduces the incidence and prevalence of pneumonia in children in low-income countries where deficiency is common. Evidence is graded Limited.
- Type 2 diabetes and HIV: Evidence that zinc supplementation prevents or improves type 2 diabetes or benefits people with HIV is graded Insufficient by the ODS.

Drug Interactions to Know
If you take any medications, the ODS identifies several clinically relevant interactions with zinc that are worth discussing with a clinician or pharmacist before starting a supplement:
- Quinolone and tetracycline antibiotics (such as ciprofloxacin): zinc can inhibit absorption of both the antibiotic and the zinc itself. Take the antibiotic at least 2 hours before or 4 to 6 hours after the zinc supplement.
- Penicillamine (used for rheumatoid arthritis and Wilson disease): zinc can reduce its absorption and action. Take zinc and penicillamine at least 1 hour apart.
- Thiazide diuretics such as chlorthalidone and hydrochlorothiazide increase urinary zinc excretion, which can decrease serum zinc concentrations over time.
- Iron supplements containing 25 mg or more of elemental iron taken at the same time as zinc can reduce zinc absorption and plasma zinc concentrations.
People with Wilson's disease must avoid extra copper entirely, which is a separate but related consideration when evaluating any mineral supplement.
Practical Takeaways
The 40 mg per day UL for zinc exists precisely because of the copper-depletion risk, and that threshold is based on ODS and National Academies Dietary Reference Intake data, not manufacturer recommendations. A few points worth keeping in mind:
- Short-term use of zinc lozenges at doses near or modestly above the UL for cold relief carries a different risk profile than sustained daily supplementation at high doses.
- Long-term supplementation programs that include zinc, such as AREDS formulas for AMD, are specifically designed to include copper alongside zinc to counteract this interaction.
- If you are considering zinc doses above 40 mg per day for any reason, the ODS notes the UL does not apply when a physician is supervising medical treatment, but that supervision is the key factor.
