Muscle Building Supplements: Why Creatine Leads and Vitamin D Supports
Creatine has the strongest evidence for boosting strength and power, while vitamin D plays an indirect but meaningful supporting role in musculoskeletal health.

If you are serious about resistance training, creatine is the supplement with the most consistent research support for improving strength and high-intensity performance. Vitamin D does not build muscle directly, but keeping levels adequate protects the bones and neuromuscular function that make training possible in the first place.
What Creatine Does in the Body
Your muscles rely on adenosine triphosphate (ATP) as their immediate fuel source during short, intense efforts. Creatine helps regenerate ATP by supplying phosphate through phosphocreatine, the compound stored in muscle tissue. According to the NIH Office of Dietary Supplements (ODS), creatine increases muscle stores of phosphocreatine used to generate ATP at the start of intense exercise, and it also speeds the re-synthesis of phosphocreatine after each effort. This is why creatine is most relevant to activities like weightlifting and sprinting rather than long-duration endurance work.
What the Research Shows for Creatine
The ODS grades the evidence for creatine in the following way:
- Strong evidence: Creatine helps supply muscles with energy for short-term, predominantly anaerobic activity by regenerating ATP through phosphocreatine.
- Strong evidence: Creatine supplementation increases strength and power and improves performance in repeated short bursts of high-intensity, intermittent exercise such as sprinting and weightlifting.
- Moderate evidence: Over weeks or months, creatine aids training adaptations to increased workloads during structured resistance training.
- Insufficient evidence: Creatine improves endurance-sport performance such as distance running or swimming. The research here does not yet support a clear benefit.
In plain terms, creatine is well-supported for the kind of training most people associate with muscle building. Its benefits for endurance sports remain unclear.
Creatine Safety and Practical Considerations
The ODS states that creatine is considered safe for short-term use by healthy adults, and evidence shows that use over several years is also safe. No adult target intake or upper limit has been formally established by dietary reference intake authorities. The most commonly reported effect is weight gain from water retention, which is expected and not harmful for most people. Anecdotal reports include nausea, diarrhea, gastrointestinal distress, muscle cramps, muscle stiffness with reduced range of motion, and heat intolerance. Staying well hydrated may help minimize these uncommon risks.
Because creatine is metabolized to creatinine and cleared by the kidneys, people with impaired kidney function should exercise particular caution. The ODS exercise and athletic performance fact sheet does not list specific well-established drug interactions for creatine, but it is prudent to speak with a clinician or pharmacist before combining creatine with nephrotoxic medications or if you have any kidney concerns. Always consult a doctor before adding any supplement to your routine, especially if you take prescription medications.
What Vitamin D Does in the Body
Vitamin D is not a performance supplement in the direct sense, but its physiological roles are closely tied to the structures that support training. According to the ODS, vitamin D promotes calcium absorption in the gut and maintains adequate serum calcium and phosphate concentrations to enable normal bone mineralization. It also prevents hypocalcemic tetany, which refers to involuntary muscle contractions, cramps, and spasms that occur when blood calcium falls too low. Additionally, vitamin D supports bone growth and remodeling by osteoblasts and osteoclasts, and together with calcium it helps protect older adults from osteoporosis.
What the Research Shows for Vitamin D
The ODS grades the evidence for vitamin D as follows:
- Strong evidence: Vitamin D promotes intestinal calcium absorption and maintains serum calcium and phosphate for normal bone mineralization.
- Strong evidence: Vitamin D deficiency causes rickets in children and osteomalacia in adults.
- Moderate evidence: Vitamin D plus calcium supplements slightly increase bone mineral density in some postmenopausal women and older men, and can reduce fracture rates in institutionalized older people.
- Limited evidence: Vitamin D supplements reduce falls or fractures in community-dwelling older adults.
- Insufficient evidence: Vitamin D supplements prevent or treat cancer, cardiovascular disease, depression, multiple sclerosis, or type 2 diabetes, or aid weight loss.
For active adults, the practical takeaway is that adequate vitamin D supports the skeletal and neuromuscular infrastructure that resistance training depends on. It is not a muscle-building agent on its own.
Vitamin D Intake Targets and Safety
According to the NIH ODS and Dietary Reference Intake data, the adult target for vitamin D is 15 mcg (600 IU) per day, rising to 20 mcg (800 IU) for adults aged 71 and older. The Tolerable Upper Intake Level from all sources, including food, beverages, and supplements, is 100 mcg (4,000 IU) per day for everyone aged 9 and older, including during pregnancy and lactation. Staying within this upper limit avoids the risk of hypercalcemia, which is an abnormally high level of calcium in the blood. A health care provider may prescribe higher doses in specific clinical situations, which is why personal dosing decisions should always involve a clinician.
Vitamin D Drug Interactions
The ODS identifies several interactions worth knowing about. Talk to your doctor or pharmacist before taking vitamin D supplements if you use any of the following:
- Orlistat (Xenical, alli): This weight-loss drug can reduce absorption of vitamin D from food and supplements.
- Statins such as atorvastatin (Lipitor), lovastatin (Altoprev, Mevacor), and simvastatin (FloLipid, Zocor): High-dose vitamin D supplements may interfere with how well these medications work.
- Corticosteroids such as prednisone (Deltasone, Rayos, Sterapred): These can lower blood levels of vitamin D.
- Thiazide diuretics such as chlorthalidone (Hygroton), indapamide (Lozol), and hydrochlorothiazide (Microzide): When combined with vitamin D supplements, these drugs can raise blood calcium to an unsafe level.
How These Two Supplements Fit Together
Creatine and vitamin D serve different purposes. Creatine directly fuels the anaerobic energy system that drives heavy lifting and sprint-style efforts, with strong evidence behind it. Vitamin D maintains the calcium balance and bone integrity that allow you to train consistently over the long term. Neither replaces a balanced diet, adequate protein intake, or a well-designed training program.
